Phospholipids

Phospholipids drive the formation of encapsulation, provide structure, and prevent the premature breakdown of LNPs. Recent studies have found that tuning the phospholipid component of an LNP can enhance delivery of nucleic acids by increasing endosomal escape in the case of phosphoethanolamine species; whereas, zwitterionic phospholipids may aid in the solubilization of nucleic acids inside the LNP1.

DOPC

DOPC, also known as 18:1 (Δ9-Cis) PC, is a phospholipid that contains unsaturated oleic acid (18:1) at the sn-1 and sn-2 positions. DOPC is a neutral helper lipid that has been shown to promote more stable laminar structures in LNPs and other drug delivery systems.

DOPE

DOPE, also known as 18:1 (Δ9-Cis) PE, is a phospholipid that contains unsaturated oleic acid at the sn-1 and sn-2 positions, a melting temperature of ~30 °C and a conical shape. DOPE tends to adopt an inverted hexagonal H(II) phase, which destabilizes endosomal membranes and facilitates endosomal escape of lipid nanoparticles.

DSPC

DSPC, also known as 18:0 PC, is a phospholipid that contains saturated stearic acid at the sn-1 and sn-2 position. DSPC is the most common phospholipid used in lipid nanoparticles (LNP) due to its high transition temperature (55C) which works to prevent the premature breakdown of the LNP in biological systems.
