ROLE OF SURFACTANTS IN MIGRATING SURFACE-INDUCED DESTABILIZATION OF LIPID NANOPARTICLES

Lipid nanoparticles (LNPs) are essential carriers for improving the stability and delivery efficiency of labile biologics, including mRNA-based therapeutics. Despite their utility, LNPs exhibit significant vulnerability to stresses that can compromise their structural integrity, resulting in cargo leakage and particle fusion. Analogous to protein formulations, LNPs are particularly prone to interfacial stresses - most notably at air-liquid and ice-liquid interfaces - which further exacerbate their instability. Although certain specialized surfactants have shown promise in reducing interfacial destabilization, surfactant-based strategies alone are insufficient to protect LNPs during freeze–thawing. Collectively, these observations highlight the pronounced sensitivity of LNPs to interfacial perturbations and emphasize the critical importance of excipient selection in preserving nanoparticle stability under diverse stress conditions.
