High Purity Excipients and Lipids in Enabling Advanced Drug Delivery

This presentation examines the impact of excipient purity on formulation performance across small molecules, proteins, and nucleic acids. A high-purity dimethyl isosorbide enhances dermal permeation with improved stability in topical small molecule systems. Case studies reveal the role of polysorbate in mitigating surface-induced protein destabilization, and the advantage of high purity surfactants in improving drug stability. Purified surfactants also stabilize RNA-loaded lipid nanoparticles against physical stress by preventing particle leakage and aggregation.
Objectives:
- Understand how excipient purity influences stability along with permeation and bioavailability in topical drug delivery.
- Investigate the impact of surfactant purity on formulation stability and strategies to mitigate enzymatic degradation
- Assess the role of purified surfactants in stabilizing LNP formulations for nucleic acid delivery upon mechanical shaking and freeze-thawing.
